No they don't. They don't have to be short haired either. Tomboys are called such because they act more like boys than like girls. Usually that means being athletic, playing outside, wearing boy's clothes, talking in a brash, masculine way (more noticeable in Japanese which has stronger gendered speech), or generally being "tough".
A common way to make a girl look like a tomboy is to give her a short haircut, a tan, and put her in shorts and a tanktop. That's almost universal costume for tomboys.
Ryuuko never seemed very boyish to me, aside from acting like a violent delinquent, but in this day and age that's not really a gendered role.
She still dresses like a girl and acts like a girl in most situations. She's really tough, but being tough alone isn't enough to make a girl a tomboy. She lacks a lot of the key aesthetic points.
